Abstract

Kantung semar (Nepenthes sp.) merupakan salah satu tanaman hias yang sering dijumpai di kawasan hutan. Tanaman ini masuk Convention on International Trade of Endangered Species (CITES), di mana semua tanaman yang masuk dalam CITES dilarang untuk diperdagangkan karena dianggap hampir punah. Untuk itu perlu ditemukan alternatif penggadaan bibit dengan pemanfaatan bioteknologi seperti kultur jaringan. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian zat pengatur tumbuh ekstrak kecambah kacang hijau pada media MS terhadap pertumbuhan planlet tanaman kantung semar secara in vitro. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) yaitu Konsentrasi Ekstrak Kecambah Kacang Hijau yang terdiri dari empat taraf (0 mL/L, 20 mL/L, 40 mL/L, 60 mL/L) sehingga terdapat 4 perlakuan dengan 3 ulangan. Uji statistika yang digunakan berupa uji non parametrik Kruskal-Wallis dan uji lanjutan menggunakan uji Mann-Whitney, dengan taraf signifikasi 5%. Hasil penelitian menunjukkan bahwa pemberian ZPT ekstrak kecambah kacang hijau dengan konsentrasi 20 ml/L mempengaruhi penambahan rata-rata tinggi tanaman sebanyak 1 cm. rata-rata jumlah daun pada konsentrasi 40 ml/L berpengaruh signifikan dan berbeda nyata dengan rata-rata jumlah daun sebanyak 9,33 tunas, sedangkan rata-rata jumlah tunas sebanyak 10,33 tunas dan rata-rata waktu muncul tunas sebanyak 3,33 tunas.

Furthermore, 40 mL/L was significantly effective in increasing the average number of leaves to 9.33, while the average number of shoots reached 10.33.
