Kesehatan dan kekebalan tubuh merupakan aspek penting yang perlu diperhatikan, terutama di era pandemi saat ini. Kemampuan tubuh dalam melawan benda asing (patogen) dapat diukur melalui titer antibodi. Penelitian ini bertujuan untuk menguji potensi ekstrak buah juwet (Syzygium cumini) sebagai imunomodulator terhadap titer antibodi tikus jantan yang diinduksi sel darah merah sapi. Penelitian ini merupakan penelitian eksperimental dengan menggunakan Rancangan Acak Lengkap (RAL) yang terdiri atas 4 kelompok dan 5 ulangan. Variabel bebas yang digunakan adalah ekstrak buah juwet (Syzygium cumini) dengan tiga dosis, yaitu 50 mg/kg BB, 100 mg/kg BB, dan 150 mg/kg BB, serta perlakuan Na CMC sebagai kontrol. Perlakuan dimulai pada hari ke-1 dan diberikan satu kali sehari selama 11 hari. Tiap kelompok hewan percobaan diinjeksikan secara intraperitoneal pada hari ke-1 dengan 0,5 ml sel darah merah sapi (SDMS) 2% dalam PBS sebagai antigen. Kontrol negatif yang digunakan adalah pemberian Na CMC. Hasil pengukuran menunjukkan bahwa nilai titer antibodi tertinggi terdapat pada kelompok perlakuan ekstrak etanol buah juwet dengan dosis 150 mg/kg BB, yaitu sebesar 2,98, diikuti oleh dosis 100 mg/kg BB dengan nilai titer antibodi sebesar 2,94, dan dosis 50 mg/kg BB.
