Abstract

ABSTRACT Patchouli oil (Pogostemon cablin Benth.) is an essential oil derived from the leaves of the patchouli plant. This oil is recognized for its primary component, patchouli alcohol, an antioxidant. The antioxidant properties of patchouli oil effectively inhibit free radical oxidation through the exchange of hydrogen atoms, forming more stable compounds. This study seeks to evaluate the antioxidant potential of patchouli oil through the FRAP method, utilizing a UV-Vis spectrophotometer for measurement at a wavelength of 692 nm. The findings indicate a regression equation of y = 0.0258x + 0.0256, with correlation coefficient of r = 0.9981 and an antioxidant activity measured at 1.7831 mgQE/g extract. Keywords: Antioxidant; Patchouli oil (Pogostemon cablin Benth.); FRAP method; UV-Vis spectrophotometer ABSTRAK Minyak nilam (Pogostemon cablin Benth.) merupakan minyak atsiri yang diperoleh dari daun tanaman nilam. Minyak ini dikenal memiliki kandungan utama patchouli alcohol, yang berfungsi sebagai antioksidan. Aktivitas antioksidan minyak nilam dapat menghambat oksidasi radikal bebas dengan cara menyumbangkan atom hidrogen, sehingga menghasilkan senyawa yang lebih stabil. Penelitian ini bertujuan untuk mengetahui potensi minyak nilam sebagai antioksidan menggunakan metode FRAP, kemudian diukur menggunakan spektrofotmeter UV-Vis pada panjang gelombang 692 nm. Dari penelitian ini diperoleh persamaan regresi y = 0,0258x + 0,0256, r = 0,9981 dan aktivitas antioksidan sebesar 1,7831 mgQE/g ekstrak.   Kata kunci: Antioksidan; Minyak nilam (Pogostemon cablin Benth.); Metode FRAP; Spektrofotometer UV-Vis

This study presents a focused investigation into the antioxidant potential of patchouli oil (Pogostemon cablin Benth.), an essential oil known to contain patchouli alcohol, a compound with recognized antioxidant properties. Given the escalating interest in natural sources for combating oxidative stress, this research is timely and relevant, contributing to the characterization of botanically derived compounds. The authors appropriately selected the Ferric Reducing Antioxidant Power (FRAP) method to evaluate the oil's capacity to inhibit free radical oxidation, a standard and reliable spectrophotometric assay. The experimental setup, utilizing a UV-Vis spectrophotometer at a wavelength of 692 nm, suggests a well-defined and reproducible analytical approach. The research successfully generated a calibration curve essential for quantifying antioxidant activity, indicated by a strong regression equation (y = 0.0258x + 0.0256) and an excellent correlation coefficient (r = 0.9981). This high 'r' value underscores the reliability and linearity of the chosen method, providing confidence in the quantitative results. The antioxidant activity of patchouli oil was subsequently determined to be 1.7831 mgQE/g extract. Expressing the activity in milligram Quercetin Equivalents per gram of extract is a common and useful metric, allowing for potential comparison with other antioxidants or natural extracts once a broader context is established. While the study provides a clear quantitative assessment of patchouli oil's antioxidant activity via the FRAP method, its overall impact could be strengthened by offering more comparative context for the reported value of 1.7831 mgQE/g extract. Without benchmarks against known synthetic antioxidants, other essential oils, or even different patchouli oil samples, the significance of this specific activity level remains somewhat isolated. Future work would benefit from exploring the activity using complementary antioxidant assays (e.g., DPPH, ABTS) to provide a more comprehensive profile, investigating the influence of extraction methods on antioxidant capacity, and potentially delving into the *in vivo* implications or specific applications where this antioxidant property could be advantageous. Nevertheless, this work lays a solid foundation by rigorously quantifying the FRAP-based antioxidant potential of patchouli oil.
